Tolerance Titans is a free online immune tolerance game where you deploy regulatory defenses to stop your own army from attacking the body. This immunology strategy game is built on the paradox at the heart of immunity: power without control is autoimmune disease.

Immune tolerance is learned, not assumed. T-cells passing through the thymus get deleted if they react to self, a filter called central tolerance, while survivors that slip out are policed by regulatory T-cells, the Tregs that patrol tissues suppressing friendly fire through peripheral tolerance. Macrophages and checkpoints all balance attack against restraint.

When tolerance fails, the immune system becomes the enemy. Type 1 diabetes, multiple sclerosis and lupus are all broken-tolerance diseases while cancer exploits the same brakes to hide. Therapies from immunosuppressants to checkpoint inhibitors work by tuning the exact dial you manage in this game.
